New HIV Vaccine Shows Unprecedented Success in Preclinical Study
A germline-targeting vaccine developed by LJI and Scripps Research generates broadly neutralizing antibodies in primates — the best HIV-fighting antibody response ever seen in animals. Human trials are underway.
Scientists at La Jolla Institute for Immunology (LJI) and Scripps Research have achieved what may be the most significant HIV vaccine breakthrough in decades: a vaccine that generated broadly neutralizing antibodies in primates at levels never seen before.
The approach, called "germline targeting," works by activating naive B cells — immune cells in their earliest, untrained state — and then shepherding them through a series of booster shots that guide their maturation. Think of it as bootcamp for the immune system, teaching B cells to recognize HIV's most vulnerable structural features and lock onto them.
The results, published in Nature, are striking. Around 44% of vaccinated rhesus macaques produced broadly neutralizing antibodies in their blood — the exact kind seen in the rare humans whose immune systems naturally fight HIV effectively.
"We succeeded in taking ultra-rare antibody responses and turning them into common responses," said LJI Professor Shane Crotty, who co-led the 14-year research effort.
HIV has evaded vaccine efforts for decades because it mutates rapidly, disguises itself with a sugar-molecule cloak, and changes shape when infecting cells — giving B cells almost no time to develop effective antibodies. The germline-targeting strategy sidesteps these defenses by training B cells on antigen models before they ever encounter the real virus.
The priming immunogen has already been tested in the HVTN 144 human trial, and a Phase 1 trial (IAVI G004) is currently underway. Plans are advancing for a full human clinical study of the complete immunization regimen.
